Measurement of Serum TSH concentration by Enzyme-linked Immunosorbent Assay



Abstract
Determination of TSH in serum is helpful in diagnosis and follow up the course of various thyroid state. Radioimmunoassay (RIA) can be regarded very sensitive method for determination of lower serum TSH concentration.
Recently, TSH-ELISA is introduced for the quantitative determination of TSH in vitro by sandwitch technique, which has several advantage over the RIA;
1. there is no radioactive material and hence no precautions need to be taken in
this regard,
2. the reagents may have a longer self-life with proper care, and
3. the enzymes used are easily determined in the usual clinical laboratory.
The authors used Enzymum-test" TSH (Boehringer mannheim) KIT for ELISA
and HTSH RIABEAD¢ç (Abott) kit for RIA and then obtained some results. The results are summarized as follows;
1. There are no significant difference between the values of ELISA and RIA. (p <0.005)
2. The regression lines and coefficient of correlation for comparison of correlationship between the values of ELISA and RIA are as follows; Y = 1. 007X - 0. 064, r=0.986, n=60.
3. The coefficients of variation were 6.3101o in intra-assay, and 4.08% in inter-assay.
